Can you simply swap the wheels & tires from an '03 GS 430 (currently has stock 17") on to a '00 GS 400 (currently has stock 16")?

There is a certain level of tolerance in the speedo/odo/... already. Lexus does not tune the car to the different available wheel sizes so they are already expecting some variance.

Like the deadmilenko already mentioned, this variance is about 1% which translates to less than 1mph on the speedo.

Just do it. :D
